Human MHC Class II (HLA-DP, DQ & DR) molecules are expressed on dendritic cells, monocytes, macrophages, and myeloid cells. MHC Class II molecules are heterodimers of polymorphic transmembrane a and b chains and generally bind exogenously derived peptides of about 10-17 amino acids. MHC Class II molecules interact with the T cell receptor on CD4+ T cells.

| Flow Cytometry Protocol | 1. Five x 10e5 cultured human Daudi cells were washed and pre incubated 5 minutes with 20ul of 250ug/ml human IgG (To block non specific binding). |
2. After which they were incubated for 45 minutes on ice with 80ul of M3887-55B at 1ug/ml.
3. Cells were washed twice and incubated with 2º reagent S7973-88P: Streptavidin/R-PE after which they were washed three times, fixed and analyzed by FACS.
4. Cells stained positive with a mean shift of 2.40 log10 fluorescent units when compared to a Mouse IgG1/Biotin negative control at a similar concentration.
5. Binding was blocked when pre incubated 10 minutes with 20ul of 0.5mg/ml M3887-55A: MHC Class 2 (beta chain).
