| MDM2, a 55kD nuclear phosphoprotein, is a Human homolog of mouse mdm2. It is E3-ubiquitin ligase consisting of a central Zinc-finger domain and a C-terminal RING finger motif. Mdm2 is a p53-binding protein and requires p53 for its transcription. The primary function of mdm2 is to recognize and bind to N-terminal transactivation domain of p53, leading to its ubiquitination, export out of the nucleus and subsequent degradation of p53 by the proteasome. Mdm2 and p53 are the members of a negative feedback control loop that regulate p53-mediated cell growth and apoptosis. MDM2 also affects cell cycle, apoptosis, and tumorigenesis through interactions with other proteins including Rb1 and ribosomal protein L5.
