Clone Host Grade Applications
Monoclonal Mouse Affinity Purified E
Mucins are heavily glycosylated glycoproteins, which constitute the major components of mucus which cover the surface of epithelial tissues. Nine distinct epithelial mucin genes (Muc-1, 2, 3, 4, 5AC, 5B, 6,7 and 8) have been identified. Various immunohistochemical and in situ hybridization studies have shown that these mucins are differentially expressed in epithelia with cell-type specificity. Muc-5AC and Muc-6 glycoproteins are the first two mucins found in superficial epithelium and Muc-6 glycoprotein is found in the deep glands. Belongs to monoclonal cluster 1-4 according to ISOMB TD-4 classification.
Catalog #M4700-05
ApplicationsSuitable for use in ELISA tests for detection and measurements of unglycosylated and underglycosylated tumor-associated MUC1 mucins in human serum. Also for use in solid-phase reagents for MUC1 isoforms capture (a) in conjunction with the same monoclonal enzyme-conjugated versions (detection of unglycosylated MUC1), or (b) with enzyme-conjugated monoclonal specific to carbohydrate portions of MUC1 (detection of underglycosylated MUC1). Other applications have not been tested.
Recommended DilutionOptimal dilutions to be determined by the researcher.
HybridomaHybridization of X63-Ag8-653 myeloma cells with spleen cells from Balb/c mice.
Affinity Constant>1x10e8
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

FormSupplied as a liquid in PBS, pH 7.4, 0.1% sodium azide, before the addition of glycerol to 40%.
Purity>90%. Purified by chromatography on protein A Sepharose. Purity is tested by electrophoresis.
ImmunogenHigh molecular weight (>300kDa) glycoprotein from human milk-fat globule molecule.
SpecificityBinds with different regions of GVTSAPDTRPAPGSTAPPAHGVTSA syn pep spanning the one repeat of VNTR extracellular potion of MUC1 molecule. Reacts with VNTR5 and VNTR20 recombinant unglycosylated fragments of MUC1 protein and underglycosilated MUC1 prepared from tumor fluids or as a result of chemical treatment of human milk MUC1. Does not recognize natural MUC1 protein isolated from human milk by affinity purification on carbohydrate epitope specific MAB. No crossreacttion with egg white avidin.
