| Mucins are heavily glycosylated glycoproteins, which constitute the major components of mucus which cover the surface of epithelial tissues. Nine distinct epithelial mucin genes (Muc-1, 2, 3, 4, 5AC, 5B, 6,7 and 8) have been identified. Various immunohistochemical and in situ hybridization studies have shown that these mucins are differentially expressed in epithelia with cell-type specificity. Muc-5AC and Muc-6 glycoproteins are the first two mucins found in superficial epithelium and Muc-6 glycoprotein is found in the deep glands. | | | Catalog # | M4700-06U | | Applications | Suitable for use in ELISA. Other applications not tested. | | Recommended Dilutions | ELISA: May be used as solid-phase reagents (5-10ug/ml) for MUC1 isoforms capture in conjunction with the same Monoclonal enzyme-conjugated versions for detection of unglycosylated MUC1, or with M4700-07 which is specific to carbohydrate portions of MUC1 for detection of underglycosylated MUC1. | | Optimal dilutions to be determined by the researcher. | | Hybridoma | X63-Ag8.653 myeloma cells with spleen cells from Balb/c mice | | Affinity Constant | >1x10e8L/mole | | Storage and Stability | May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG | | Clone No | 10J46 | | Host | Mouse | | Source | Human | | Concentration | ~1mg/ml | | Form | Supplied as a liquid in PBS, pH 7.4, 0.1% sodium azide. | | | Purity | Purified by Protein A affinity chromatography. (>90% by SDS-PAGE) | | Immunogen | High molecular weight (>300kD) glycoprotein from human milk-fat globule molecule | | Specificity | Recognizes a region of GVTSAPDTRPAPGSTAPPAHGVTSA synthetic peptide spanning the one repeat of VNTR extracellular portion of human MUC1 molecule. Reacts with VNTR5 and VNTR20 recombinant unglycosylated fragments of MUC1 protein and underglycosylated MUC1 prepared from tumor fluids or as a result of chemical treatment of human milk MUC1. Does not recognize natural MUC1 protein isolated from human milk by affinity purification on carbohydrate epitope specific monoclonals and belongs to Monoclonal Cluster 1-4 according to ISOMB TD-4 classification. Does not cross-react with egg white avidin. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. | | Alternate names | CA 15-3, CD227 Antigen, Mucin 1 Cell Surface Associated |
