| Mucins are heavily glycosylated glycoproteins, which constitute the major components of mucus which cover the surface of epithelial tissues. Nine distinct epithelial mucin genes (Muc-1, 2, 3, 4, 5AC, 5B, 6,7 and 8) have been identified. Various immunohistochemical and in situ hybridization studies have shown that these mucins are differentially expressed in epithelia with cell-type specificity. Muc-5AC and Muc-6 glycoproteins are the first two mucins found in superficial epithelium and Muc-6 glycoprotein is found in the deep glands. | | | Catalog # | M4700-07 | | Applications | Suitable for use in ELISA and Immunohistochemistry. Other applications have not been tested. | | Recommended Dilutions | ELISA: Suitable for the detection of under-glycosylated tumor-associated MUC1 mucins in human serum. | | Immunohistochemistry (FFPE and Frozen): 50ug/ml or less | | Optimal dilutions to be determined by the researcher. | | Affinity Constant | >1x10e8 | | Hybridoma | X63-Ag8-653 myeloma cells with spleen cells from BALB/c mice | | Recommended ELISA pairs | Capture: M4700-08 or M4700-05 | | Detection: M4700-07 | | Storage and Stability | May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG | | Clone No | 1.B.877 | | Host | Mouse | | Source | Human | | Concentration | ~5mg/ml | | Form | Supplied as a liquid in PBS, pH 7.4, 0.1% sodium azide. | | Purity | Purified by Protein A affinity chromatography. | | Immunogen | High molecular weight (>300kD) glycoprotein from human milk-fat globule molecule | | Specificity | Recognizes underglycosylated and natural MUC1 protein (isolated from human milk by affinity chromatography). Does not crossreact with normal serum proteins and human tissues of nonepithelial origin. Binds with high efficiency with different epitopes within the VNTR tandem repeat peptide conformations and peptide- carbohydrate compositions. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
