Mucins are heavily glycosylated glycoproteins, which constitute the major components of mucus which cover the surface of epithelial tissues. Nine distinct epithelial mucin genes (Muc-1, 2, 3, 4, 5AC, 5B, 6,7 and 8) have been identified. Various immunohistochemical and in situ hybridization studies have shown that these mucins are differentially expressed in epithelia with cell-type specificity. Muc-5AC and Muc-6 glycoproteins are the first two mucins found in superficial epithelium and Muc-6 glycoprotein is found in the deep glands.

ApplicationsSuitable for use in ELISA and Immunohistochemistry. Other applications not tested.
Recommended DilutionELISA: 5-10ug/ml for MUC1 capture in conjunction with the same monoclonal enzyme-conjugated versions, or with M4700-07 specific to carbohydrate portions of MUC1 (detection).
Immunohistochemistry: Frozen sections or neutral-buffered formalin-fixed paraffin-embedded sections.
Optimal dilutions to be determined by the researcher.

Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

SpecificityRecognizes human Mucin-1. Binds with high efficiency with an epitope within the VNTR tandem repeat peptide region of MUC1 molecule. Reacts with VNTR20 recombinant unglycosylated fragment of MUC1 protein (and very weakly with monomeric MUC1 peptide), with deglycosylated and underglycosylated natural MUC1 (but not with completely glycosylated natural milk MUC1). Belongs to MAb clusters 6 and 7 according to ISOMB TD-4 classification. Non crossreactive with normal serum proteins and human tissues of nonepithelial origin.
