| Neurofilament proteins (NFPs) are a macromolecular complex comprised of 3 polypeptides designated as NF-L, NF-M and NF-H. NFPs are found in the perikarya, particularly in neuronal axons throughout the central and peripheral nervous system. Since NFPs are major structural proteins and biochemically quite stable, antibodies to NFPs are useful probes in studies of neuronal expression, morphology, connectivity and pathology. The presence or absence of NFP in a variety of nervous system or neuroendocrine tumors can provide useful information about the original cell type of the tumor. In addition, the normal NFP staining pattern is altered in a variety of human diseases including Alzheimer’s disease, diffuse cortical Lewy body disease, Parkinson’s disease and amyotrophic lateral sclerosis (Lou Gehrig’ disease). Alterations in the NFP expression pattern are also seen in most toxin-induced, sporadic and heriditary axonopathies occurring in humans and animals. | | | Catalog # | N2160-04E | | Applications | Suitable for use in Flow Cytometry, ELISA, Western Blotting, Immunoprecipitation, Immunohistochemistry. Other applications not tested. | | Recommended Dilution | Flow Cytometry: | | ELISA: 0.1–0.5ug/ml | | Immunohistochemistry: 5–10ug/ml (paraffin-embedded or frozen sections) | | Immunoprecipitation: 2–5ug | | Immunoblotting(1): 0.5–1.0ug/ml | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG1,k | | Clone No | 3G204 | | Host | Mouse | | Source | Rat | | Concentration | 0.5mg/ml | | Form | Supplied as a liquid in PBS, pH 7.2, 0.1% sodium azide. | | Purity | Purified | | Immunogen | Rat neurofilaments | | Specificity | Reacts with the 160 kD polypeptides of human neurofilament. It specifically recognize a phosphateindependent epitope in the rod (core) domain of NF-M.(1,2) Reactivity with lampey NF-M has also been shown. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
