Neurogranin is a 78 amino acid neuron-specific substrate of protein kinase C that accumulates predominantly in dendritic shafts and spines of neurons. Immunoreactive neurogranin has been detected in many brain regions but is particularly abundant in the mammaliam forebrain. Immunostaining is completely abolished by pre-adsorption of the antibody with the immunogen peptide.

Recommended DilutionImmunoblot Analysis: 1:500-1:5000 of this lot detected phospho-Neurogranin/Neuromodulin in RIPA lysates from rat brain extract which had been phosphorylated by 500ng of Protein Kinase C.
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

ImmunogenKLH-conjugated, synthetic peptide corresponding to aa35-47 (ATKIQA[pS]FRGHIT) of rat neuromodulin. The homologous sequence in neurogranin contains the phosphorylation site at Ser36.
SpecificityRecognizes phosphorylated Neurogranin, Mr ~7-17kD and Neuromodulin, Mr ~43-45kD. A non-specific protein was also detected, Mr ~55kD. Species Crossreactivity: Rat and mouse. Predicted to crossreact with bovine and human based on sequence homology.
