Several subtypes of NPY Receptors have been described of which the cloned Y1-, Y2- and Y5-receptors, members of the superfamily of G-protein coupled receptors, appear to be the endogenous binding sites for NPY. The Y(2) receptor is the predominant Neuropeptide Y (NPY) receptor subtype in the brain. Y(2) receptor-mediated effects have been reported principally based on the observations using the C-terminal fragments of NPY. Recent studies indicate an involvement of the receptor in food intake, gastrointestinal motility, cardiovascular regulation, and neuronal excitability.

ApplicationsSuitable for use in ELISA, Immunohistochemistry and Immunofluorescence. Other applications not tested.
Recommended DilutionImmunohistochemistry: 1:500-1:1000; Used on perfusion fixed tissue. Perfusion: 1) calcium-free Tyrode’s solution, 2) paraformaldehyde-picric acid fixative and 3) 10% sucrose in PBS. Tissues were dissected and stored overnight in 10% sucrose, PBS.
Immunofluorescence (Indirect): Incubate slides with blocking buffer for 1 hour at RT. Dilute N2176-63G with blocking buffer. Remove blocking buffer from slide and incubate with N2176-63G for 18-24 hours at 4°C. Rinse slides 3X and then incubate with appropriate secondary antibody for 1 hour at RT. Rinse slides again 3X and set coverslip. Examine staining using fluorescence microscopy.
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
