Niemann-Pick C1-like 1 (NPC1L1) is a transmembrane protein that plays a critical role in cholesterol absorption (1). It is highly expressed in small intestine and localized along the brush border in both human and mouse (2,3). NPC1L1 mediates cholesterol uptake via its vesicular endocytosis (4). Ezetimibe, a potent cholesterol absorption inhibitor that is being clinically used to treat hypercholesterolemia (5), prevents NPC1L1 from incorporating into clathrin-coated vesicles and thus inhibits cholesterol uptake (4).

Applications
Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested.
Recommended Dilution
Western Blot: 1:1000, incubate membrane with diluted primary antibody in 5% BSA, 1X TBS, 0.1% Tween 20 at 4°C with gentle shaking, overnight. Immunoprecipitation: 1:50 Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Synthetic peptide corresponding to central residues of human NPC1L1. Uniprot Accession: Q9UHC9

Supplied as a liquid in 10mM sodium HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol.

Purified by Protein A and peptide affinity chromatography.
Specificity
Recognizes endogenous levels of total human NPC1L1.
