Nitric oxide (NO) and reactive oxygen species (ROS) are important mediators to produce harmful or protective functions. NO can react with superoxide anions (O2-), yielding the toxic oxidizing agent peroxynitrite (ONOO-). Peroxynitrite induces nitration of tyrosine residues leading to changes of protein structure and function, and alterations in signaling pathways. Antibodies to nitrotyrosine have been used to monitor the status of peroxynitrite-induced modification of proteins.

ApplicationsSuitable for use in ELISA and Western Blot. Other applications not tested.
Recommended DilutionWestern Blot: 1-5ug/ml. Proteins extracted from human A431 cells or other cells pretreated with peroxynitrite for 5 min may serve as good positive control.
ELISA: 1:10,000-1:50,000 using 50-100ng N2700-10 Nitrated MW Standards, Western Blot/well.
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
