| This is an N-ras oncogene encoding a membrane protein that shuttles between the Golgi apparatus and the plasma membrane. This shuttling is regulated through palmitoylation and depalmitoylation by the ZDHHC9-GOLGA7 complex. The encoded protein, which has intrinsic GTPase activity, is activated to a GTP-bound form by a GTPase activating protein and inactivated to a GDP-bound form by a guanine nucleotide-exchange factor. Defects in this gene are a cause of juvenile myelomonocytic leukemia (JMML). | | | Catalog # | N5385-01D | | Applications | Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. | | Recommended Dilutions | Western Blot: 0.01-0.03ug/ml. A band at ~21kD is seen with human lysates in A431 cell line. In transfected HEK293 transiently expressing NRAS bands of ~21kD and 25kD are seen. These bands are not seen in non-transfected HEK293. | | Immunohistochemistry (Paraffin): 2-3ug/ml. Shows staining of cytoplasm and membranes of enterocytes in paraffin embedded human small intestine. | | Optimal dilutions to be determined by the researcher. | | Positive Control | Human lysates of cell line A431 | | Storage and Stability | May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Goat | | Source | Human | | Concentration | ~0.5mg/ml | | Form | Supplied as a liquid in Tris saline, pH 7.3, 0.5% BSA, 0.02% sodium azide | | Purity | Purified by immunoaffinity chromatography. | | Immunogen | Synthetic peptide corresponding to C-RMKKLNSSDDGTQ, from the internal region of the protein sequence according to NP_002515.1. | | Specificity | Recognizes human NRAS.
