Specifications

Neurotrophin-3 (NT3) is a member of the neurotrophin family, that controls survival and differentiation of mammalian neurons. This protein is closely related to both nerve growth factor and brain-derived neurotrophic factor. It seems to promote the survival of visceral and proprioceptive sensory neurons. The mature peptide of this protein is identical in all mammals examined including human, porcine, rat and mouse. NT3 is a secreted protein that is expressed in brain and peripheral tissues.

Biological. ActivityThe ED50 of NT3 is typically 0.1-0.2ng/ml as measured in a cell proliferation assay using a TrkB transfected PC12 cell line.
Applications
Suitable for use in Western Blot. Other applications not tested.
Recommended DilutionsWestern Blot: The NT3 protein migrates at ~14-15kD in SDS-PAGE and has a predicted molecular mass of 13.6kD. NT3 separates into a number of isoforms with a pI from 6.6 to at least 10 in 2D PAGE due to post-translational modifications. The unmodified NT3 has a predicted pI of 9.2.
Optimal dilutions to be determined by the researcher.
Storage and StabilityLyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
