| Nucleophosmin (NPM), a nucleocytoplasmic shuttling protein with prominent nucleolar localization, regulates the ARF-p53 tumor-suppressor pathway. Translocations involving the NPM gene cause cytoplasmic dislocation of the NPM protein (1). NPM is a major nucleolar protein which is 20 times more abundant in tumor or proliferating cells than in normal resting cells. Data suggest that transcription factor YY1 may play a role in NPM gene expression (2). A chromosomal translocation associated with myelodysplastic syndrome and acute myeloid leukemia (AML) was found to rearrange part of the nucleophosmin (NPM) gene. Myeloid leukemia factor 1 (MLF1) binds NPM and studies have indicated that MLF1 is normally located in the cytoplasm, whereas NPM-MLF1 is targeted to the nucleus, with highest levels in the nucleolus.
