| The p27Kip1 (p27) gene encodes an inducible inhibitor of cyclin-dependent kinase activity. It associates with cyclin E-Cdk2 complexes in vivo and in vitro, prevents their activation, and inhibits previously activated complexes, and its overexpression obstructs cell entry into S phase. It potently inhibits Rb phosphorylation by cyclin E-Cdk2, cyclin A-Cdk2, and cyclin D2-Cdk4. It is highly conserved and broadly expressed in Human tissues, with highest levels in skeletal muscle, lowest in liver and kidney and its mRNA levels are similar in proliferating and quiescent cells. It has a region of sequence similarity to p21Cip1/WAF1, the Cdk inhibitor whose transcription is stimulated by p53. The protein levels are high during G0 and G1 phases of the cell cycle, resulting in cyclin/CDK inhibition. It maps to 12p13.1-p12 region of Human chromosome.
