| MAGUKs are scaffold proteins that interact with cytoskeleton and regulate cell proliferation, signaling pathways and intracellular junctions and associate in tumor suppression and receptor clustering. PALS1, a 675aa protein, is a member of p55-like subfamily of MAGUKs. It is a human ortholog to Drosophila Stardust and mouse homolog of protein associated with LIN7. It consists of central SH3 domain, C-terminal PDZ and GUK domains, and an L27domain. PALS1 is targeted to tight junction with its L27 domain and plays a significant role in tight-junction protein targeting, vesicle trafficking and establishment and maintenance of cell polarity in polarized cells like mammalian neurons and epithelial cells. In Drosophila, PALS1 interacts with PATJ and PAR6 binding sites and forms 2 evolutionary conserved multi-protein complexes viz., Crumbs-PALS1-PATJ and Cdc42-Par6-Par3, implicated in the assembly of tight junctions.
