| L1 is a major capsid protein of type 16 human papilloma virus. Infection with specific types of HPV has been associated with an increased risk of developing cervical neoplasia. HPV types 6 and 11 have been associated with relatively benign diseases such as genital warts but types 16 and 18 are strongly associated with cervical, vaginal, and vulvar malignancies. | | | Catalog # | P3105-08 | | Applications | Suitable for use in Immunoprecipitation, Western Blot, Immunohistochemistry, Immunocytochemistry and Immunofluorescence. Other applications not tested. | | Recommended Dilutions | Immunohistochemistry: reacts with fixed tissue. Dilute with buffer pH 7.5-8 to desired working volumes. For extensive dilution, protein containing or other stabilizing medium should be used. | | Western Blot: Detects a protein of 41kD in addition to HPV-16 L1 at 57kD. The 41kD protein was observed in a wide range of uninfected mammalian cells. Exhaustive recloning failed to eliminate this specificity. The specificity is seen only in Western Blot. No reactivity is observed by Immunoprecipitation or Immunohistochemical detection. This unwanted specificity might be due to crossreactivity between HPV-16 L1 and a cryptic epitope in mammalian cells that is revealed only under denaturation (i.e. by boiling in detergent and reducing agent). | | Immunofluorescence: use cells fixed in acetone or paraformaldehyde and immunoperoxidase staining of paraffin sections. | | Optimal dilution to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months.
