| The poly (ADP-ribose) polymerase (PARP) is involved in cell recovery from DNA damage, such as methylation of N3-adenine, which activates the base excision repair process. PARP [Poly (ADP-ribose) polymerase] is a 116kD nuclear chromatin-associated enzyme that is cleaved during apoptosis by caspase-3 into a 24kD fragment containing the DNA binding domain and an 89kD fragment containing the catalytic and automodification domains. The 24kD-fragment irreversibly bind to DNA and may contribute to the irreversibility of apoptosis by blocking the access of DNA repair enzymes to DNA strand breaks. | | | Catalog # | P3113-07J | | Applications | Suitable for use in Western Blot and Flow Cytometry. Other applications not tested. | | Recommended Dilution | Western Blot: 1-2ug/ml | | Flow Cytometry (intracellular): 1-3ug per test | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG | | Clone No | 4H226 | | Host | Mouse | | Source | Human | | Concentration | ~0.5mg/ml | | Form | Supplied as a liquid in PBS containing 0.2% gelatin and 0.05% sodium azide. | | Purity | Purified by Protein G affinity chromatography. | | Immunogen | Synthetic peptide containing amino acids near 214/215-cleavage site of human PARP. | | Specificity | Recognizes human PARP, Cleaved (Poly ADP-Ribose Polymerase). Specifically detects a 89-kDa cleavage fragment containing the catalytic and auto modification domain. | | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
