| Paxillin can be diffusely distributed in the cytoplasm, co-localized with actin at the cell periphery, or aggregated at focal adhesions. Paxillin has several motifs which allow its interaction with other proteins. These motifs include LD motifs, LIM domains, an SH3 domain binding site, and SH2 domain binding sites. Paxillin complexes with proteins of focal adhesions including vinculin, talin, with the kinases FAK, PYK2, Src, and Csk, with ARF GTPase activating proteins, and with the adaptor protein v-Crk. Paxillin is phosphorylated at several residues, including tyrosine 31, tyrosine 118, tyrosine 181. The phosphorylation of paxillin appears to be important for several biological functions including S-phase transition and neurite extension. |

| Catalog # | P3113-90 |
| Applications | Suitable for use in ELISA, Western Blot, Immunoprecipitation, and Immunocytochemistry. Other applications not tested. |
| Recommended Dilution | Western Blot: 0.5-2ug/ml detects paxillin in RIPA lysates from 3T3 fibroblasts using 20ug per lane for minigels. |
| Immunoprecipitation | 4ug per 500ug of cell lysate. |
| Immunocytochemistry | 5-10ug/ml (gave positive immunostaining for paxillin in REF (rat embryo fibroblast) 52 cells fixed with 3.7% paraformaldehyde for 5 minutes). |
| Optimal dilutions to be determined by the researcher. |
| Positive Antigen Control | Add 2.5ul 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for immunoblot analysis. For use as a positive control for antibodies. |
| Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C or colder. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
