Of the 11 families of phosphodiesterase (PDEs) identified, PDE4, PDE7, and PDE8 are specific for cAMP. However, isoforms of the PDE4 family often account for most of the cAMP-hydrolyzing activity of a cell. Four genes (PDE4A, B, C and D) make up the PDE4 family. One or more subtype variants of PDE4B are ubiquitously present in all mammalian cells. In the CNS, PDE4B subtype variants are expressed in varying ratios. Peripheral tissues exhibit differential expression of PDE4B1, B2 and B4 variants.
Applications
Suitable for use in Immunoprecipitation and Western Blot. Other applications have not been tested.
Recommended Dilutions
Immunoprecipitation: 7ug/ml Western Blot: 1-3ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic peptide derived from the N-terminal region of human and rat PDE4B protein.
Form
Supplied as a liquid in PBS, pH 7.4, 0.09% sodium azide.
Purity
Purified by immunoaffinity chromatography.
Specificity
Reacts with the human and rat PDE4B protein. On Western blots, it identifies a single band at ~83kD. Bands of unknown origin may be observed at ~160 and ~260kD. Reactivities have been confirmed with NG96 human neuroblastoma lysate and rat brain homogenate.
