Clone Host Grade Applications
Monoclonal Mouse Affinity Purified E B IP
Phosphorylation and dephosphorylation of cellular proteins are central steps in transducing extracellular signals to the cell nucleus. Phosphorylated epitopes may serve as docking sites for the assembly of protein complexes or may alter the 3-dimensional protein structure thus modulating enzymatic activity or the ability to undergo protein-protein-interactions. Modification of proteins on serine residues is mediated by serine/threonine kinases.
Catalog #P4076-17
ApplicationsSuitable for use in ELISA, Immunoprecipitation and Western Blot. Other applications have not been tested.
Recommended DilutionELISA: 0.05ug/ml
Immunoprecipitation: 1-10ug per 10e6 pervanadate-treated A431 cells
Western Blot: 1ug/ml for AP/BCIP/NBT (MTT) detection; 0.1ug/ml for HRP/ECL detection. BSA/Tween 20 based blocking buffer. Do NOT use milk/casein for blocking and dilution.
Optimal dilutions to be determined by the researcher.
Positive Control IncludedP4076-22A: Phosphoserine/Phosphothreonine, Positive Control
Supplied as a liquid in PBS, 10mM NaF, PEG, sucrose, 0.09% sodium azde. Add 75ul SDS-PAGE sample buffer and incubate at 90°C for 5 minutes.
Western Blot: 20ul/lane for HRPO/ECL detection. 5ul/lane for HRPO/DAB detection.
Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Clone TypeMonoclonal
Clone No6D659
FormSupplied as a liquid in PBS, 0.09% sodium azide, PEG, and sucrose.
PurityPurified from serum-free cell culture supernatant by subsequent thiophilic adsorption and size exclusion chromatography.
ImmunogenPhosphoserine, conjugated to KLH.
SpecificityRecognizes a broad range of serine-phosphorylated proteins in crude cell extracts, requiring either proline or lysine C-terminal to phosphoserine. Please note that phosphoserine detection by monoclonal antibodies is always dependent on the surrounding amino acid sequence. Species Crossreactivity: Human, mouse, rat, canine
