| Protein phosphorylation is a complex biochemical process that ultimately influences the mechanism by which a cell responds to a particular stimulus. A class of enzymes referred to as protein kinases regulates this process; these enzymes catalyze the transfer of the gama-P of ATP to a recipient protein that acts as a substrate. This post-translational modification is primarily directed onto serine and threonine amino acid residues within a protein. It has been noted that changes in the phosphorylation status of these phosphoproteins can alter the biological function of a protein. Additionally, aberrations in signal transduction cascade have been linked to a number of diseases including cancer, diabetes, heart disease, inflammation and neurological disorders. | | | Catalog # | P4077 | | Applications | Suitable for use in Western Blot, ELISA, and Immunoprecipitation. Other applications have not been tested. | | Recommended Dilution | Western Blot: 2ug/ml; Mouse brain lysate. Incubate at RT for 4 hrs. | | ELISA: 0.5ug/ml (kinase assay). Binding of Ly-pT-Gly-HRP | | (0.2ug/100ul/well) to an anti-phosphothreonine coated microplate (0.5ug/100ul/well) is inhibited by 95% with the addition of 2-10ug/ml anti-phosphothreonine. No inhibition is evident by inhibition of phosphoserine, phosphotyrosine or threonine at concentrations of up to 1mg/ml. | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Mouse | | Concentration | ~0.25mg/ml | | Form | Supplied as a liquid in PBS, pH 7, 0.01% sodium azide, 50% glycerol. | | Purity | Purified by immunoaffinity chromatography. | | Immunogen | Phosphothreonine conjugated to KLH and phosvitin. | | Specificity | Recognizes phosphothreonine, peptidyl-phosphothreonine and threonine-phosphorylated proteins.
