Phosphorylation and dephosphorylation of cellular proteins are central steps in transducing extracellular signals to the cell nucleus. Phosphorylated epitopes may serve as docking sites for the assembly of protein complexes or may alter the 3-dimensional protein structure thus modulating enzymatic activity or the ability to undergo protein-protein-interactions. Modification of proteins on serine residues is mediated by serine/threonine kinases.

Applications
	Suitable for use in ELISA, Immunoprecipitation and Western Blot. Other applications not tested.
Recommended Dilutions	Western Blot: 1ug/ml for HRPO/ECL detection. Recommended blocking buffer: BSA/Tween 20 based blocking buffer. Do not use milk or casein for blocking.
	Immunoprecipitation: 1-10ug per 10e6 pervanadate-treated A431 cells.
	ELISA: 0.05ug/ml
	Optmal dilutions to be determined by the researcher.
Positive Control Included	P4076-22A: Phosphoserine/Phosphothreonine, Positive Control
	Add 100ul 2x SDS-PAGE sample buffer and incubate at 90°C for 5minutes.
	Western Blot: 20ul/lane HRPO/ECL
Storage and Stability	May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
