| PIAS1 (protein inhibitor of activated STAT) proteins blocks the DNA binding activity of Stat1 and inhibits Stat1-mediated gene activation in response to cytokine stimulation (1). PIAS1 also plays a crucial role as a transcriptional coregulator in the p53 pathway and the steroid hormone signaling pathway and functions as an E3 type small ubiquitin-like modifier (SUMO) ligas, stabilizing the interaction between UBEZ1 and the substrate, and as a SUMO-tethering factor.
