| Biological Activity | 5ng/ml of this lot of PDGF-BB supported a 1.9-fold increase in cell growth of NIH 3T3 broblasts over control cultures without PDGF-BB. The optimal PDGF concentration in cell culture can vary depending on cell type. |
| PDGF Growth Assay | 1. Prepare a cell suspension in 2% platelet poor serum to a nal concentration of 1x10e4 cells/ml. Aliquot 4ml/tube to seed quadruplicate wells (1ml/well) of test, positive and negative controls. |
| 2. Prepare PDGF-BB following instructions above for a concentration of approximately 1ug/ml and appropriate dilutions. |
| 3. Add PDGF-BB directly to the 4ml suspension, mix well and dispense 1ml/well ( nal concentration 0-10ng/ml). |
| 4. Refeed on day 3 with growth medium + PDGF-BB. |
| 5. Count and record cell numbers on day 5. Calculate the stimulation factor over negative control results. |
| Storage and Stability | Lyophilized powder may be stored at -20°C. May be stored at 4°C for short-term only. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
