Specifications

PDGF is a mitogenic peptide growth hormone carried in the alpha-granules of platelets. It is released when platelets adhere to traumatized tissues. Connective tissue cells near the traumatized region respond by initiating the process of replication. The synthesis of PDGF can be induced by Interleukin-1, Interleukin-6, TNF-alpha, TGF-beta and EGF.
Catalog #P4258-15D
Recombinant Human Platelet Derived Growth Factor-BB is a homodimeric, glycosylated, polypeptide chain containing 109 amino acids and having a molecular mass of 32,021 Daltons.
Endotoxin<0.1ng/ug (1EU/ug)
Dimers and Aggregates<1% as determined by silver-stained SDS-PAGE gel analysis.
Biological ActivityPDGF-BB is fully biologically active when compared to standard. The ED50, calculated by the dose-dependent proliferation of murine BALB/c 3T3 indicator cells (measured by 3H-thymidine uptake) is <1ng/ml, corresponding to a Specific Activity of 1MIU/mg.
Protein ContentProtein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm using the absorbency value of 0.55 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a standard solution of PDGF-BB as a Reference Standard.
SequenceThe sequence of the first five N-terminal amino acids was determined and was found to be Ser-Leu-Gly-Ser-Leu.
Storage and StabilityLyophilized powder may be stored at 4°C for short-term only. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
SourceRecombinant, Human expressed in Saccharomyces cerevisiae
Purity95% by RP-HPLC or reducing/non-reducing SDS-PAGE Silver Stain. Chromatographically purified.
FormSupplied as a lyophilized powder in 20mM sodium acetate buffer, pH 7.4. Reconstitute in sterile dH2O 0.1mg/ml. Can be further diluted to other aqueous solutions.
Important NoteThis product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological.
