Peripheral myelin protein 22 (PMP22) is a small, hydrophobic a tetraspan glycoprotein, with proposed roles in peripheral nerve myelin formation, cell-cell interactions, and cell proliferation. It is most prominently expressed by Schwann cells as a component of compact myelin of the peripheral nervous system (PNS), thus representing 2 to 5% of all myelin proteins. It is also observed in nonneural cells and tissues during development, such as in the epithelial cells of the lung and intestine. The human PMP-22 gene has been mapped to chromosome 17p11.2 and codes for a 22kD transmembrane glycoprotein. It plays a crucial role in normal physiological and pathological processes in the peripheral nervous system which includes correct myelination during development of peripheral nerves, the stability of myelin, and the maintenance of axons. Recent study revealed that genetic alterations in the PMP22 gene including duplications, deletions, and point mutations are responsible for the most common forms of hereditary peripheral neuropathies including Charcot-Marie-Tooth disease type 1A (CMT1A), hereditary neuropathy with liability to pressure palsies (HNPP), and a subtype of Dejerine-Sottas Syndrome (DSS).
