PEDF R (Pigment epithelium derived factor receptor; also ATGL, PNPLA2 and desnutrin) is a 55kD member of the PNPLA family of proteins. It is expressed in adipocytes, hepatocytes, skeletal muscle and pigment epithelium. PEDF R specifically hydrolyses triglycerides, releasing long-chain fatty acids. Mouse PEDF R is described as being a 486aa type II transmembrane
(TM) protein. Its TM segment is stated to span aa9-29. Alternatively, it may have a hydrophobic lipid-association domain between aa 267-295. It contains a palatin domain (aa10-179) with a conserved GxSxG enzymatic motif. There are multiple splice forms. Two show alternate start sites at Met57 and Met232, while two others show a deletion of aa163-196 and 253-308,
respectively. Over aa 62-253, mouse PEDF R shares 98% and 93% aa identity with rat and human PEDF R, respectively.

Recommended DilutionPeptide ELISA: 1:128,000.
Western Blot: 0.03-0.1ug/ml. Approx 60kD band observed in Human Heart lysates (calculated MW of 55.3kD according to NP_065109.1). Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Peptide BlockingCorresponding peptide is available for Peptide Blocking studies. See P4350-15D-P. Antibody is typically 0.5mg/ml and peptide is supplied as a 100ul pellet. When peptide is reconstituted in 200ul water, the concentration would also be 0.5mg/ml. To start, the best ratio would be 1:1 (molar excess of peptide relative to antibody when identical volumes are mixed). Mix equal volumes of peptide and antibody at the required dilution and leave at ambient temperature. It is best is to have two identical blots to be incubated with equal amount of antibody, but one with the antibody pre-adsorbed to the peptide for 20min. Then incubate and develop the two blots in parallel.
