| Human PgR exists in two forms | 116kD (B-form) and 81kD (A-form). It acts as ligand activated transcription factor to regulate expression of the target genes. Null mutation in PgR gene leads to pleiotrophic reproductive abnormalities. Expression of PgR has been suggested to reflect a intact estrogen regulatory machinery and therefore, predict better clinical response to endocrine therapy than ER alone. |
| Applications | Suitable for use in Immunofluorescence, Immunoprecipitation, Western Blot, Immunohistochemistry and Sedimentation in Sucrose Gradient. Other applications have not been tested. |
| Recommended Dilution | Immunoprecipitation (Use Protein G): 2ug/mg protein lysate |
| Western Blot: 1-2ug/ml for 2hrs at RT |
| Immunohistochemistry (Formalin/paraffin): 1:50-1:100 for 60 min at RT. Staining of formalin/paraffin tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min. |
| Optimal dilutions to be determined by the researcher. |
| Positive Control | T47D or breast Carcinoma. |
| Storage and Stability | May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
