| Prohibitin is an evolutionarily conserved gene with homologues found in organisms ranging from yeast to man. The gene was first identified in resting, mammalian cells. It has antiproliferative activity and is also involved in diverse processes as normal cell cycle regulation, replicative senescence and cellular immortalization. The 30kD protein has been located in the inner membrane of mitochondria, where it is postsynthetically modified, as well as on the plasma membrane of B cells, where it is associated with the IgM receptor. The protein is also found in the nucleus, where it has been reported to bind the retinoblastoma protein and function as a novel regulator of E2F activity. Mutations in the prohibitin gene cause sporadic breast tumors.
