Protein Gene Product 9.5 (PGP9.5) is an abundant protein in many tissues, but especially so in neurons where it has been effectively used as a phenotypic marker and is immunologically distinct from neuron specific enolase. The 27kD protein was first identified by high resolution two dimensional PAGE. Standard immunohistochemical techniques have demonstrated the presence of PGP9.5 in neurons and nerve fibers at all levels of the central and peripheral nervous system, in many neuroendocrine cells, in segments of the renal tubules, in spermatogonia and Leydig cells of the testis, in ova and in some cells of both the pregnant and non-pregnant corpus luteum. PGP9.5 is known to be a member of the ubiquitin C- terminal hydrolase family and immunohistochemical studies have shown that the protein is enriched in several ubiquinated inclusion bodies, suggesting that such structures may be metabolically dynamic regions of the cell.

ApplicationsSuitable for use in Western Blot, ELISA, and Immunohistochemistry. Other applications have not been tested.
Recommended DilutionsWestern Blot: 1:1000 detected Protein Gene Product 9.5 on 10ug of mouse brain lysates. Mouse brain lysate was resolved by electrophoresis, transferred to PVDF and probed with P9102-71. Proteins were visulaized using a donkey/goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.
Immunohistochemistry (Paraffin): 1:50-1:500. Antigen retrieval using citrate, pH 6 is required.
Optimal dilutions to be determined by the researcher.

Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
