Based on PTEN’s structural features, the PTEN protein was originally thought to be a protein tyrosine phosphatase, however, recent evidence indicates that PTEN protein is actually a lipid phosphatase. The target substrate for PTEN is phosphatidylinositol-3, 4, 5-triphosphate (Ptd- Ins(3,4,5,)P3). Ptd-Ins(3,4,5,) P3 is a critical second messenger involved in cell signaling and growth control (1-4). PTEN functions to “turn off” the Ptd-Ins(3,4,5,)P3 pathway and thereby suppress cellular proliferation. Immunostaining study showed epithelium of prostate gland, some normal breast ductal epithelium, lower crypt of normal gastric mucosa and some scattered glial cells stained PTEN positively in normal tissue sections. Normal and hypertrophied prostate stained strongly. No staining or diminished staining was shown in prostate carcinoma and breast carcinoma in tissue sections, which indicated the deletion or mutation of PTEN gene (13-16). This result implies that this antibody may play a role for clinical cancer research, especially prostate carcinoma. Further studies are necessary for PTEN protein expression in other types of human tumors.

Positive Control Normal human prostate tissue, NIH 3T3 cells, K562 cells, MCF-7 cells and rat 1 cells overexpressing a tagged human PTEN protein.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
