Pituitary tumor-transforming gene 1 (PTTG-1) is an oncogene that is expressed in normal tissues containing proliferating cells and in tumors. PTTG is downstream of the MAP kinase cascade and its phosphorylation at Ser162 by MAPK in vitro. The phosphorylation plays an essential role in PTTG’s transactivation function. PTTG has a DNA-binding domain, and it activates c-Myc (an oncogene) transcription by binding to the promoter in a protein complex. Studies found that expression of human PTTG correlates with cell proliferation in a cell cycle-dependent manner and the association of hPTTG with Ku, the regulatory subunit of DNA-dependent protein kinase, is disrupted when a DNA double-strand breaks occurs.

ApplicationsSuitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended DilutionELISA: 0.1-1ug/ml
Western Blot: 2-4ug/ml. Recognizes only the 28kD band in MCF-7 cell lysate.
Immunohistochemistry: 1-2ug/ml in formalin-fixed, paraffin embedded tissues. For best results heat induced epitope retrieval (HIER) with citrate buffer, pH 6.0, is required prior to staining.
Optimal dilutions to be determined by the researcher.

Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
