Rab3A is a 25kD GTP-binding protein implicated in neurotransmitter release (1). The geranylgeranyl groups at the carboxyl-terminus of Rab3A are essential for its association with synaptic vesicles, guanine nucleotide-dissociation factor (GDI), and for GTP binding (2). Rab3A is believed to regulate vesicle fusion through a cycle of GDP/GTP exchange and GTP hydrolysis, which is in turn modulated by GTPase activating protein (GAP), GDI, guanine nucleotide-releasing factor (GRF), and rabphilin (3). The dif-ferent guanine nucleotide binding states of Rab3A is postulated to affect its ability to associate or dissociate with membranes during synaptic vesicle recycling. Its GTP-bound form, which represents the active form of Rab3A, associates with membrane and regulates vesicle docking and fusion. Rab3A and its homolog, Rab3C, have been shown to be co-localized to and dissoci-ated from synaptic vesicles after the stimulation of exocytosis (3). Deletion of the Rab3A structural gene has minimal effect on the morphology of the nervous system and spontaneous and evoked synaptic currents (4). However, Rab3A knockout mice exhibited that an increased rate of synaptic depression after repeated stimulation, suggesting Rab3A is involved in maintaining a steady supply of vesicles for release.
