| Rab proteins are low-molecular-weight GTP-binding proteins that form the largest branch of the Ras superfamily of GTPases. Located on the cytoplasmic face of organelles and vesicles, rab proteins are involved in intracellular membrane fusion reactions. Three membrane proteins, synaptosomal associated protein of 25kD (SNAP-25), synaptobrevin, and syntaxin, form the core of a ubiquitous membrane fusion machine that interacts with the soluble proteins N-ethylmaleimide-sensitive factor (NSF) and a-SNAP. Rab proteins, in coordination with the core fusion machinery and Munc-18, help to mediate vesicle docking and fusion. There exist over 40 Rab proteins that have been described in mammals. Rab 11 is a known marker for protein trafficking, sorting, and recycling in the endosomal pathway. Rab11 is a member of the YPT-1 subfamily of Ras-related GTP-binding proteins. Rab11 mRNA has been detected in a variety of tissues including brain, testis, spleen, heart and gastrointestinal mucosa. In the gastric fundus the Rab11 protein is expressed in parietal cell tubulovesicles membranes. Expression of the Rab11 protein appears to occur in discrete epithelial cell populations where it localizes to apical vesicular populations. | | | Catalog # | R0009-03 | | Applications | Suitable for use in Western Blot. Other applications not tested. | | Recommended Dilution | Western Blot: 5ug/ml. Detects a ~22kD protein representing Rab 11 in CV1, PC3, and HeLa cell | | extract. Another less prominent, unknown band is also detected by this antibody at ~35kD. | | Optimal dilutions to be determined by the researcher. | | positive control | HEK293 cells | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | ~0.5mg/ml | | Form | Supplied as a liquid in PBS, pH 7.4, 0.05% sodium azide, before the addition of glycerol to 40%. | | Purity | Purified by immunoaffinity chromatography. | | Immunogen | Synthetic peptide corresponding to aa1-17, MGTRDDEYDYLFKVVLIC, from human Rab11. | | Specificity | Recognizes human Rab11a protein at ~25kD.
