RANKL and RANK are members of the TNF superfamily of ligands and receptors that play an important role in the regulation of specific immunity and bone turnover. RANK (receptor) was originally identified as a dendritic-cell- membrane protein, which by interacting with RANKL augments the ability of dendritic cells to stimulate naïve T-cell proliferation in a mixed lymphocyte reaction, to promote the survival of RANK + T cells, and to regulate T-cell-dependent immune response. RANKL, which is expressed in a variety of cells including osteoblasts, fibroblasts, activated T-cells and bone marrow stromal cells, is also capable of interacting with a decoy receptor called OPG. Binding of soluble OPG to sRANKL inhibits osteoclastogenesis by interrupting the signaling between stromal cells and osteoclastic progenitor cells, thereby leading to excess accumulation of bone and cartilage.

ApplicationsSuitable for use in Immunohistochemistry, ELISA and Western Blot. Other applications have not been tested.
Recommended Dilutions
ELISA (Sandwich, Indirect): 0.5-2.0ug/ml Use 100ul/well antibody solution. Allows the detection of 0.2-0.4ng/well of recombinant hsRANKL.
Western Blot: 0.1-0.2ug/ml. Detection limit for recombinant hsRANKL is 1.5-3.0ng/lane, under either reducing or non-reducing conditions.
Immunohistochemistry: 0.25ug/ml on formalin-fixed, paraffin-embedded sections of human metastatic carcinoma of lymph node from breast tissue. Incubate overnight at 4ºC. Heat induced antigen retrieval with a pH 6.0 sodium citrate buffer is recommended.
Optimal dilutions to be determined by the researcher.
Storage and StabilityLyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute by adding sterile 40-50% glycerol, ddH2O. Aliquot and store at -20°C. Reconstituted product is stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
