Rb2/p130, a member of the retinoblastoma family of growth and tumor suppressor genes, is extensively implicated in the control of cell cycle and differentiation.The protein, acts as a negative regulator of cell cycle progression, blocking the cells in the G, phase and are also crucial effectors in embryonic development as well as in differentiation. It is a phosphoprotein that mainly acts by modulating the function of several nuclear transcription factors, such as those of the E2F family. It is upregulated at late differentiation stages in neuroblastoma cells and its overexpression induces differentiation in neuroblastoma cells. The protein recruits and targets histone methyltransferases SUV420H1 and SUV420H2, leading to epigenetic transcriptional repression. The gene contains 22 exons spanning over 50 kb of genomic DNA. It has been mapped to human chromosome16q12.2, an area in which deletions have been found in several human neoplasias including breast, ovarian, hepatic, and prostatic cancers which is in support of an involvement of the p130/RB2 gene in human cancer as a tumor suppressor gene. It also forms a complex with Rad50 through RINT-1 and blocks telomerase-independent telomere lengthening in normal cells.
