| RhoGAP1 is a 439aa intracellular GTPase activator for the Rho proteins (Rho, Rac and Cdc42). It has a proline-rich domain, Src homology 3 (SH3) binding domain and three consensus box regions characteristic of RhoGAP. It stabilizes charges developing on the guanine nucleotide during the transition state for GTP hydrolysis and thus contributes to GAP-stimulated catalysis and converts the Rho proteins to their putatively inactive GDP-bound state with Cdc42 being the preferred substrate. Along with Rho proteins they modulate cytoskeletal changes and cell signaling. It is known to regulate the basal apoptosis of cells through the JNK pathway and also play a crucial role in regulating Rho-mediated signaling pathways.
