Specifications

Purified from an E.coli strain carrying a T4 RNA ligase overproducing plasmid. Catalyses the ATP-dependent intra- and intermolecular formation of phosphodiester bonds between 5'-phosphate and 3'-hydroxyl termini of single-stranded RNA or DNA. The minimal substrate is a nucleoside 3', 5'-diphosphate.

Unit Definition	1 unit of enzyme catalyses the conversion of 1 nanomole of 5'-[32P]-(rA)12-18 to a phosphatase-resistant form in 30 minutes at 37°C.
Activity Assay	50mM Tris-HCl (pH 7.5), 10mM MgCl2, 10mM DTT, 1mM ATP, 10uM 5'-[32P]-(rA)12-18 (10uM in 5'-termini).
Storage Buffer	10mM Tris-HCl (pH 7.5), 1mM DTT, 50mM KCl, 0.1mM EDTA and 50% glycerol.

Storage and Stability
May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 24 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
