SB 203580 is a highly specific inhibitor of SAPK2a/p38 (IC50=600nM). It suppresses the activation of MAPKAP Kinase-2 and MAPKAP Kinase-3. SB 203580 does not significantly inhibit SAPK/JNK or Erk/MAP kinases at 100mM. It inhibits IL-1 and TNF-a production from LPS-stimulated human monocytes and the human monocyte cell line THP-1. SB 203580 has been shown to inhibit activation of the HIV-1 long terminal repeat by IL-1 and TNF-a. It has also been shown to be an effective inhibitor of inflammatory cytokine production in vivo in mice and rats.

Applications Suitable for use in Protein Kinase Inhibition Assay. Other applications not tested.
Recommended Dilutions Protein Kinase Inhibition Assay: A 20 minute pre-incubation of immunoprecipitated SAPK2a/p38 MAPK with 1-10uM of SAPK3/p38 Inhibitor (SB203580) inhibits the kinase activity 80-95%. Optimal concentration of SB 203580 needed may depend on the amount of enzyme used.
Storage and StabilityLyophilized powder may be stored at 4°C for short-term only. For Long term storage, store at -20°C. Lyophilized powder is stable for 36 months at -20°C. Reconstitute to nominal volume by adding sterile DMSO, aliquot and store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
