| Smurf1 (Smad ubiquitination regulatory factor-1) is a member of the Hect family of proteins, which also includes the ubiquitin (Ub) E3-type ligases Nedd3 and E6-AP. E3 ligases are involved in the enzymatic reactions of the Ub conjugating pathway that targets proteins for degradation by the 26S proteasome. In the Ub pathway, E3 ligases catalyze the transfer of Ub from Ub-conjugating enzymes to the individual protein substrate. As an E3 ligase, Smurf1 preferentially promotes the transfer of Ub to Smad proteins, thereby targeting them for degradation. Smurf1 regulates the inhibitory activity of Smad by targeting it to the plasma membrane, and is also known to enhance the degradation of the inhibitory Smad1 and Smad5, thereby down-regulating the BMP pathway, which controls embryomic development and the specification of ectoderm- and mesoderm-derived tissues. | | | Catalog # | S1014-60 | | Applications | Suitable for use in Western Blot. Other applications have not been tested. | | Recommended Dilution | Western Blot: 1-3ug/ml | | Optimal dilutions to be determined by the researcher | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Host | Rabbit | | Source | Human | | Concentration | 0.25mg/ml | | Form | Supplied as a liquid in PBS, pH 7.4, 0.1% sodium azide. | | Purity | Purified by immunoaffinity chromatography. | | Immunogen | Synthetic peptide derived from an internal region of human Smurf1 (Smad ubiquitination regulatory factor 1, Smad-specific E3 ubiquitin ligase 1, Smad-ubiquitin E3 ligase Smurf1-beta). | | Specificity | Reacts with human and mouse Smurf1. On Western blots, it identifies the target band at ~100kD. Reactivity has been confirmed with human A375, NGP96, HepG2, MeWo cell lysates, and mouse liver and testis homogenates. Based on amino acid sequence homology, reactivity with rat may also be observed. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
