Spectrin is a cytoskeletal protein of muscle fibers and labeling of spectrin is used to monitor muscle membrane integrity. This antibody should be used in combination with dystrophin antibodies to ensure the membrane is intact and that the absence of dystrophin staining is real and not due to tissue freezing or handling artifact. It is intended as a positive control for muscle membranes.

ApplicationsSuitable for use in Western Blot, Immunohistochemistry and Electorn Microscopy Gold. Other applications not tested.
Recommended DilutionWestern Blot: 1:25-1:50
Immunohistochemistry: 1:100 for 1 hr. at RT
Electorn Microscopy Gold: NEAT for 90 min. at RT Light fixation with 2% formaldehyde + 0.001% glutaraldehyde for 1 hour, 2.3M sucrose used as cryoprotec- tant is recommended.
Optimal dilutions to be determined by the researcher.
Positive Control
Western Blot: Skeletal muscle. Major band at approximately 253kD in muscle, 220kD in red blood cell “ghosts”. (Journal of Biological Chemistry. 265:20449-20454 (1990)).
Immunohistochemistry: Normal human striated muscle frozen in isopentane chilled in liquid nitrogen (staining pattern continuous rim of labeling at the periphery of muscle fibers)
Electorn Microscopy Gold: Normal human striated muscle frozen in isopentane chilled in liquid nitrogen (staining pattern close to cytoplasmic face of the plasma membrane of muscle fibers)
Storage and StabilityLyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
