Spinesin, also known as Transmembrane Protease, Serine 5 (TMPRSS5) is a type II transmembrane protein that includes a short N-terminal cytoplasmic tail (aa 1-49), a transmembrane domain (aa 50-70), a stem region and a serine protease domain (residues 71-457). Spinesin occurs predominately in neurons, in their axons, at the synapsis of motor neurons in the spinal cord, and in some oligodendrocytes. The amino acid sequence of human Spinesin is 94%, 82%, 81% and 79% identical to that of chimpanzee, canine, rat and mouse.

HybridomaProduced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, NS0-derived, recombinant human Spinesin (rhSpinesin; aa 71-457, Accession # NP_110397).
ApplicationsImmunoprecipitation: This antibody was used at a concentration of 25ug/ml to immunoprecipitate rhSpinesin from conditioned cell culture medium. The recovered rhSpinesin can be detected in Western blots using a polyclonal antibody.
Western Blot: This antibody can be used at 1-2ug/ml with the appropriate secondary reagents to detect human Spinesin. Using a colorimetric detection system, the detection limit for rhSpinesin is approximately 25 ng/lane under non-reducing conditions. Use of this antibody under reducing conditions is not recommended. Chemiluminescent detection will increase sensitivity by 5 to 50 fold. For Western blot under reducing conditions, AF2495 and BAF2495 are recommended.
Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect human Spinesin. The detection limit for rhSpinesin is approximately 20 ng/well.
Optimal dilutions should be determined by each laboratory for each application.
Storage and StabilityLyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
