| Stage-Specific Embryonic Antigens (SSEAs) were originally identified by monoclonal antibodies (mAb) generated against defined carbohydrate epitopes associated with lacto- and globo-series glycolipids. The SSEA-4 specific monoclonal antibody was generated from a mouse immunized with the 2102Ep human teratocarcinoma cell line. This mAb recognizes the terminal structure of the sialyl galactosylgloboside Luke (LKE) antigen expressed on human red blood cells and platelets. SSEA-4 expression has been reported on ACHN renal tubular cells and also on normal human kidney. The expression of SSEA-4 on human teratocarcinoma-derived cell lines prompted investigations into the expression of globosides during embryonal differentiation. Embryonic carcinoma cells that express SSEA-4 rapidly lose expression of SSEA-4 as the cells differentiate). In addition, human embryonic stem cells express SSEA-4. Studies have utilized this mAb for the detection of SSEA-4 on mouse oocytes through the early blastocyst stage. However, SSEA-4 has not been detected on mouse embryonic carcinoma or mouse embryonic stem cells. Finally, studies have shown that SSEA-4 is expressed on primary sensory neurons of the rat and other species.
