Synnuclein family of proteins is abundantly expressed in neuronal cytosol and presynaptic terminals. In vertebrates they are encoded by three different genes. Synucleins have been specifically implemented in three major diseases: Alzheimerís (AD), Parkinsonís (PD) and breast cancer. In AD, a peptide derived from alpha-synuclein forms an intrinsic component of plaque amyloid. In PD, an alpha-synuclein accumulates in Lewy bodies. An allele of alpha-synuclein has been linked to many familial cases of PD. In breast cancer increased expression of gamma synuclein correlates with the disease progression. Synucleins appear to be involved in the membrane plasticity in developing song control system of songbirds.

ApplicationsSuitable for use in Western Blot, Immunocytochemistry and Immunohistochemistry. Other applications not tested.
Recommended DilutionsWestern Blot: 1:5000-1:10,000 detects a- or b-Synuclein recombinant protein nitrated at Tyr39. Nitrated and unnitrated a-synuclein recombinant protein were resolved by electrophoresis, transferred to nitrocellulose and probed with S9500-28B (1:5,000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
Optimal dilutions to be determined by the researcher.

Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
