| Tau is the major microtubule-associated protein in neuronal axons that regulates microtubule stability, neurite outgrowth, and other microtubule -dependent functions. The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between both. Six tau isoforms are produced in adult Human brain by alternative mRNA splicing from a single gene. Inclusion of a 31 amino acid repeat encoded by exon 10 of the tau gene gives rise to the three isoforms with four microtubule-binding repeats each. The other three isoforms have three repeats each. Abundant neurofibrillary lesions made of the protein constitute a defining neuropathological characteristic of Alzheimer's disease.
