| Tau is a neuronal microtubule-associated protein found predominantly on axons. The function of tau is to promote tubulin polymerization and stabilize microtubules. Tau, in its hyper-phosphorylated form, is the major component of paired helical filaments (PHF), the building block of neurofibrillary lesions in Alzheimer’s disease (AD) brain. Hyper-phosphorylated tau is also found in neurofibrillary lesions in a range of other central nervous system disorders. Hyper-phosphorylation impairs the microtubule binding function of tau, resulting in the destabilization of microtubules in AD brains, ultimately leading to the degeneration of the affected neurons. Numerous serine/threonine kinases, including GSK-3b, protein kinase A (PKA), cyclin-dependent kinase 5 (cdk5) and casein kinase II, phosphorylate tau. Serine 262 can be phosphorylated by GSK-3b, PKA, CamKII and MARK and has been found to be a major site in AD brain. This antibody, combined with Tau [pS356]), enables one to distinguish between phosphorylation at the two distinct residues, rather than a combined epitope as detected by 12E8 antibody. | | | Catalog # | T1032-60 | | Applications | Suitable for use in Western Blot. Other applications not tested. | | Recommended Dilution | Western Blot: 1:1000 | | Optimal dilutions to be determined by the researcher. | | Positive Control | Recombinant human Tautreated, or untreated with PKA or African green monkey kidney (CV-1) cells, stably expressing human four repeat tau and SV40 small T antigen to induce specific inhibition of PP2A. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | Not determined | | Form | Supplied as a liquid in DPBS (without Mg2+ and Ca2+), 1mg/ml BSA (IgG, protease free), pH 7.3, 0.05% sodium azide, 50% glycerol.
