| Thrombomodulin is a 110-120kD, endothelial cell-specific type I transmembrane glycoprotein. It binds thrombin, rendering it unavailable for clotting, and activates protein C, which also serves as an anticoagulant. Mouse Thrombomodulin is 577aa in length. It contains a 510aa extracellular domain (ECD) (aa17-517) that shows one C-type lectin domain (aa31-167) and six EGF-like regions (aa240-480). Thrombin binds to aa403-480. One isoform may exist that shows a deletion of the C-terminal 86 amino acids (aa492-577). Over aa17-517, mouse Thrombomodulin is 66%, 85% and 67% aa identical to human, rat and canine TM, respectively. |

| Western Blot: 1-2ug/ml. Using a colorimetric detection system, the detection limit for recombinant mouse Thrombomodulin is ~25ng/lane under non-reducing conditions. Use under reducing conditions is not recommended. Chemiluminescent detection will increase sensitivity by 5-50 fold. |
| Immunohistochemistry: 25ug/ml detects Thrombomodulin in frozen sections of normal mouse kidney and heart. |
| Immunocytochemistry: 10ug/ml. Used with bEnd.3 cells. Cells were fixed with PBS, 4% paraformaldehyde and blocked with PBS, 10% normal donkey serum, 0.1% Triton X-100, 1% BSA. Between each step, cells were washed with PBS, BSA. |

| Storage and Stability | Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
