Clone Host Grade Applications
Monoclonal Mouse Affinity Purified B IP FC IF
Thrombospondin is a protein from platelet a-granules. It is made up of three identical subunits bound by interchain disulfides. It is secreted at sites of platelet activation and aggregation and is involved in the processes of chemotaxis, adhesion, proliferation and differentiation of leukocytes, fibroblasts, smooth muscle and endothelial cells.
Catalog #T5100-06CX
ApplicationsSuitable for use in Electron Microscopy, Flow Cytometry,
Immunofluorescence, Immunoprecipitation, Western Blot and blocks binding of TSP-1 to IAP/CD47. Not suitable for Immunohistochemistry. Other applications not tested.
Recommended DilutionImmunoprecipitation (Native verified): 2ug/mg protein lysate, Use Protein G.
Western Blot: 1-2ug/ml for 2hrs at RT, Non-reduced only.
Blocks Binding of TSP-1 to IAP/CD47: 25-50ug/ml
Optimal dilutions to be determined by the researcher.
Cellular LocalizationSecretory granules, Golgi complex, endoplasmic reticulum, extracellular matrix
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Clone TypeMonoclonal
Clone No3F352
FormSupplied as a liquid in 10mM PBS, pH 7.4. Also available with BSA and azide. See T5100-06C.
PurityPurified by Protein G affinity chromatography.
ImmunogenCa2+-replete, native puri ed human TSP from supernatant of thrombin-activated platelets. MW of Antigen
SpecificityRecognizes Thrombospondin. Shows no crossreactivity with fibronectin, fibrinogen or von Willebrand factor. It blocks TSP-mediated agglutination of fixed RBCs. It shows no effect on TSP-mediated agglutination of fixed, activated platelets. It inhibits both thrombin -and A23187-induced aggregation of washed, live (not fixed) platelets without affecting the secretion of serotonin. Inhibits adhesion of melanoma G361 cells to TSP. Blocks binding of C-terminal domain to Integrin-Associated Protein (IAP)/CD47. Species Crossreactivity: Human, bovine, porcine, equine, canine, sheep and rat.
