| Thrombospondin is a protein from platelet a-granules. It is secreted at sites of platelet activation and aggregation and is involved in the processes of chemotaxis, adhesion, proliferation and differentiation of leukocytes, fibroblasts, smooth muscle and endothelial cells. |

| Catalog # | T5100-06D |
| Applications | Suitable for use in Electron Microscopy, Flow Cytometry, Immunofluorescence, Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications have not been tested. |
| Recommended Dilution | Immunoprecipitation (Native verified) (Use Protein G): 2ug/mg protein lysate |
| Western Blot: 1-2ug/ml for 2hrs at RT |
| Immunohistochemistry (Formalin/paraffin): 1:25 to 1:50 for 60 min at RT |
| Note: Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min |
| Optimal dilutions to be determined by the researcher. |
| MW of Antigen | ~450kD (non-reduced); 170 to 180kD (reduced) |
| Epitope | Collagen Type V-binding domain of TSP |
| Positive Control | Platelets in placenta, megakaryocytes in bone marrow. |
| Cellular Localization | Secretory granules, Golgi complex, endoplasmic reticulum, extracellular matrix |
| Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |

| Purity | Purified by Protein G affinity chromatography. |
| Immunogen | Reduced and alkylated puri ed human TSP (fully denatured) from the supernatant of thrombin-activated platelets |
| Specificity | Recognizes human TSP. Inhibits TSP-collagen interaction. Its binding to TSP is unaffected by glycosaminoglycans (e.g. hyaluronic acid, chondroitin sulfate and heparin). Its binding is enhanced by EDTA i.e. at low conc. of Ca2+. It shows no crossreactivity with fibronectin, fibrinogen and von Willebrand factor. Shows a mild crossreaction with TSP 2. Species Crossreactivity: Bovine, porcine, equine, canine, sheep, mouse, and rat. |
